LRRK2 and preventive therapies in Parkinson disease

Abstract

The discovery that pathogenic mutations in the LRRK2 gene increase LRRK2 kinase activities and that small molecule LRRK2 kinase inhibitors blocking these activities can be neuroprotective in preclinical PD models have placed LRRK2 at the center of disease modification efforts in PD. Recent investigations suggest also that LRRK2 may play a role in the pathogenesis of iPD. In my talk I shall review the steps that led to the discovery that LRRK2 mutations can cause PD, and the characteristics of LRRK2-PD that are most relevant for LRRK2-targeted therapies and clinical trials. Challenges that lie ahead in implementing disease modification trials in LRRK2 mutation carriers will be discussed.
